By blacklisting tags like cartoon_network and disney.
Or by searching copytags:0
There isn't a valid tag for r34 content, but you could blacklist the most common tags in the copyright category:
https://e621.net/tags?commit=Search&search%5Bcategory%5D=3&search%5Bhide_empty%5D=1&search%5Border%5D=count
Blacklisting the big companies (like nintendo, disney, and hasbro) or franchises (like pokemon) would get a majority of the rule-34 type stuff.
As far as I know, there isn't a way to blacklist an entire category of tags. (That would also hit original content that happens to be posted here, so I don't think that'd be something you want.)

See e621:cheatsheet#post-based_metatags
You can include or exclude posts based on the number of tags in a certain category.
Also one can exclude rule 34 content by adding rating:s
